URLs with out "/" on end failing

If I request a URL without a trailing "/", the page won't load.

I.e. If request http://www.mysite.co.uk/level1/level2 after about 20-30 seconds, I get redirected to http://www.mysite.co.uk:{sites-iis-port-number}/level1/level2 (http://www.mysite.co.uk:31232/level1/level2) with "Web page is not available" error

Any ideas on what may be happening here?

I am using EPiServer CMS 6, and there is no scope to upgrade to R2 at this stage

Try using the the developer toolbar in IE (Network) to investigate what is happening. Sounds like an infinitive loop.
